EntransterTM-in vivo是英格恩生物公司(Engreen Biosystem Co,Ltd.)最新研发合成的用于动物体内转染的转染试剂。EntransterTM-in vivo动物体内转染试剂可将小片断RNA如siRNA,miRNA,mimic、inhibitor转染到动物体内,在动物体内进行RNA干扰等实验。

EntransterTM-in vivo试剂不是病毒,改变了病毒转染方法复杂、周期长、不稳定、花费大、安全性等诸多问题。EntransterTM-in vivo转染效率高,方法简单,无需病毒包装,直接将RNA送入目的细胞的细胞核中表达。

EntransterTM-in vivo,非脂质体,采用纳米技术合成,通过保护和浓缩核酸,避免核酸被体液破坏,并通过物理作用将核酸富集在细胞表面,增强核酸进入细胞组织,从而产生作用。EntransterTM-in vivo能耐受体内复杂的环境,不产生免疫反应。

方法简单

如图所示,只需要将核酸和转染试剂分别稀释,混合,无需过滤,即可直接注射动物。

转染效果

将EntransterTM-in vivo与Ambion in vivo siRNA(作用于凝血因子VII)和阴性siRNA通过尾静脉注射成年小鼠,2天后,取动物肝脏检测。在mRNA水平和蛋白水平观察干扰效果。见上图,图中最左侧组为对照组注射阴性siRNA为3mg/kg,后边3组为每kg动物注射阳性siRNA量分别为1mg/kg,2mg/kg和3mg/kg情况。

各器官转染

根据推荐用量注射EntransterTM-in vivo和siRNA(作用于Lamin A/C)和阴性siRNA到成年小鼠。注射后2天收集相应的组织,分离RNA,用qRT-PCR分析Lamin A/C基因的表达水平。图4为尾静脉结果,图5为各器官分别局部注射结果。
